Retrotransposition and cell-to-cell transfer of foamy viruses.

چکیده
A remarkable feature of the prototype foamy virus (PFV) replication pathway has been reported to consist of the ability to retrotranspose intracellularly with high efficiency (M. Heinkelein, T. Pietschmann, G. Jármy, M. Dressler, H. Imrich, J. Thurow, D. Lindemann, M. Bock, A. Moebes, J. Roy, O. Herchenröder, and A. Rethwilm, EMBO J. 19:3436-3345, 2000). PFV intracellular retrotransposition (IRT) was reported to be enhanced by coexpression of fusion-defective envelope protein. To investigate the possibility of cell-to-cell transfer of PFV genomes, which could mimic IRT, we performed cocultivation experiments with cells transfected with an IRT-competent and marker gene-expressing PFV vector together with cells expressing a different marker and measured cells positive for both markers. The findings corroborated the initial report on IRT of Env-deficient PFV. Furthermore, they indicated that viral cores that have incorporated fusion-deficient Env can be transferred from cell to cell in a cell type-specific manor. One possible explanation consists of a minor alternative cleavage site in Env that can be used to expose the fusion peptide of the Env transmembrane protein, which appears to be required for virus uptake.

Packaging cell lines for simian foamy virus type 1 vectors.
Foamy viruses are nonpathogenic retroviruses that offer several unique opportunities for gene transfer in various cell types from different species. We have previously demonstrated the utility of simian foamy virus type 1 (SFV-1) as a vector system by transient expression assay (M. Wu et al., J. Virol. 72:3451-3454, 1998). In this report, we describe the first stable packaging cell lines for fo...
متن کاملDevelopment and Cytogenetic Characterization of a Continuous Bovine Kidney Cell Line (IRKHBK) and Evaluation its Susceptibility to some Viruses
In this syudy a continuous bovine kidney cell line derived from a primary bovine kidney cells was established for the first time in Iran. The cells were originating from two-day-old normal male calf of Holstein breed. The cell cultures were continuously passaged following complete proliferation of primary cells. The specific properties or characteristics of the cell were defined using cytogenet...
متن کاملEvaluating of Iran Razi Khedmati Bovine Kidney (IRKHBK) Cell Line for Isolation and Propagation of Viruses
Abstract Background and Aims: To identify and isolation of viruses a susceptible cell is need which can be primary or line. The use of primary cells is laborious, time consuming and expensive, while the use of cell lines is much easier and faster. IRKHBK cell line was originated from primary cells of bovine kidney by continuous passage. The aim of this study was to introduce the IRKHBK cell ...
متن کاملEvolution of viruses and cells: do we need a fourth domain of life to explain the origin of eukaryotes?
The recent discovery of diverse very large viruses, such as the mimivirus, has fostered a profusion of hypotheses positing that these viruses define a new domain of life together with the three cellular ones (Archaea, Bacteria and Eucarya). It has also been speculated that they have played a key role in the origin of eukaryotes as donors of important genes or even as the structures at the origi...
متن کاملEffect of Heavy Metals on Silencing of Engineered Long Interspersed Element-1 Retrotransposon in Nondividing Neuroblastoma Cell Line
Background: L1 retrotransposons are the most active mobile DNA elements in human genome. Unregulated L1 retrotransposition may have deleterious effect by disrupting vital genes and inducing genomic instabilities. Therefore, human cells control L1 elements by silencing their activities through epigenetic mechanisms. It has been shown that cell division and heavy metals stimulate the frequency of...
